朱砂根、山豆根及其药对的HPLC指纹图谱建立及5种化学成分
的含量测定

摘 要 目的 建立朱砂根、山豆根及其药对的指纹图谱,同时测定其中5种成分的含量。方法 以水为溶剂,结合冷冻干燥技术
制备朱砂根、山豆根单煎冻干粉和朱砂根-山豆根药对合煎冻干粉样品;采用高效液相色谱(HPLC)法建立3种冻干粉样品的指纹
图谱,并同法测定没食子酸等5种成分的含量。结果 朱砂根、山豆根单煎冻干粉样品和朱砂根-山豆根药对合煎冻干粉样品分别
有5、10、14个共有峰;与对照图谱的相似度均大于0.90;朱砂根-山豆根药对合煎冻干粉样品中,指认3号峰为没食子酸、4号峰为
苦参碱、6号峰为氧化苦参碱、8号峰为岩白菜素、14号峰为三叶豆紫檀苷。朱砂根单煎冻干粉样品中,没食子酸、岩白菜素的平均
含量分别为0.499 3、4.962 6 mg/g;山豆根单煎冻干粉样品中,苦参碱、氧化苦参碱、三叶豆紫檀苷的平均含量分别为3.046 0、2.336 6、
0.278 6 mg/g;朱砂根-山豆根药对合煎冻干粉样品中,没食子酸、苦参碱、氧化苦参碱、岩白菜素、三叶豆紫檀苷的平均含量分别为
0.560 6、2.548 7、1.382 2、5.960 7、0.279 1 mg/g,转移率为8.87%~513.19%。结论 所建立指纹图谱及含量测定方法稳定、可行,可
用于朱砂根、山豆根及朱砂根-山豆根药对的质量控制;朱砂根-山豆根药对合煎冻干粉样品中苦参碱、氧化苦参碱的平均含量
降低。

ABSTRACT OBJECTIVE To establish the fingerprints of Ardisia crenata, Sophora tonkinensis and their couplet medicines, and
to determine the contents of five components in them. METHODS Using water as solvent, single lyophilized powder of A. crenata
and S. tonkinensis and combined lyophilized powder of their couplet medicines were prepared by combining lyophilization
technology. The fingerprints of three lyophilized powder samples were established by using high-performance liquid chromatography
(HPLC), and the contents of 5 kinds of components such as gallic acid were determined simultaneously. RESULTS There were 5,
10 and 14 common peaks in the fingerprints for single lyophilized powder of A. crenata and S. tonkinensis and combined
lyophilized powder of their couplet medicines; the similarities of them with the control fingerprints were all greater than 0.90. For
