马齿苋乳膏对变应性接触性皮炎小鼠皮肤瘙痒及皮肤屏障功能

          摘   要  目的  研究马齿苋乳膏对变应性接触性皮炎(ACD)小鼠皮肤瘙痒及皮肤屏障功能的影响及机制。方法  制备马齿苋提取
          液[1 g/mL(以生药量计)]含量分别为10%、20%的马齿苋低、高浓度乳膏。将60只BALB/c小鼠随机分为空白组、模型组、糠酸莫
          米松乳膏组(阳性对照)、空白基质乳膏组、马齿苋低浓度乳膏组、马齿苋高浓度乳膏组。除空白组外,各组小鼠涂抹2,4-二硝基氯
          苯溶液诱导建立ACD模型;同时空白组及模型组小鼠涂抹生理盐水,其余各组小鼠涂抹相应药物,每天1次,每次约0.5 g,连续14
          d。末次给药24 h后,观察小鼠皮肤损伤情况并评分;观察小鼠皮肤组织病理学形态变化;检测小鼠血清中免疫球蛋白E(IgE)、肿
          瘤坏死因子α(TNF-α)水平;检测小鼠背根神经节(DRG)中MAS相关G蛋白偶联受体(包括MrgprA3、MrgprC11、MrgprD)mRNA
          的表达;检测小鼠皮肤组织中皮肤屏障功能相关蛋白 Claudin-1、Occludin 的表达。结果  与空白组比较,模型组小鼠皮肤损伤
          严重,表皮结构缺失,皮肤组织角化过度,可见大量炎症细胞浸润;皮肤损伤评分,血清中 IgE、TNF-α 水平,DRG 中 MrgprA3、
          MrgprC11、MrgprD mRNA 表达水平均显著升高(P<0.05 或 P<0.01),皮肤组织中 Claudin-1、Occludin 蛋白表达水平均显著降低
         (P<0.05)。与模型组比较,马齿苋低、高浓度乳膏组小鼠皮肤损伤明显减轻,表皮增厚及真皮层海绵样变性缓解,炎症细胞浸润
          减轻,上述定量指标大部分显著逆转(P<0.05 或 P<0.01);空白基质乳膏组大鼠上述皮肤损伤、病理学形态以及定量指标无显
          著差异。结论  马齿苋能够改善 ACD 小鼠皮肤瘙痒,并修复皮肤屏障功能,其作用机制可能与下调小鼠 DRG 中 MrgprA3、
          MrgprC11、MrgprD mRNA的表达,上调皮肤组织中Claudin-1、Occludin蛋白表达相关。

          ABSTRACT    OBJECTIVE To study the effects and mechanism of Portulaca oleracea cream on skin pruritus and barrier function
          in  allergic  contact  dermatitis (ACD)  mice.  METHODS  Low-concentration  and  high-concentration  P.  oleracea  creams  were
          prepared, with the P. oleracea extract solution (1 g/mL, calculated by crude drug) concentrations of 10% and 20%. Sixty BALB/c
          mice  were  randomly  allocated  into  blank  group,  model  group,  Mometasone  furoate  cream  group (positive  control),  blank  matrix
          cream group, P. oleracea low-concentration and high-concentration cream groups. Except for blank group, ACD model was induced
          in  each  group  using  2,4-dinitrochlorobenzene  solution.  The  blank  group  and  model  groups  received  normal  saline,  while  the
          remaining  groups  were  treated  with  their  respective  creams,  once  a  day,  at  a  dose  of  approximately  0.5  g  per  application,
          continuously for 14 days. At 24 h post-final administration, skin lesions of mice were observed and scored; pathological changes of
