枳壳中3,5,6,7,8,3′,4′-七甲氧基黄酮对湿阻中焦大鼠的改善

          摘  要  目的  研究枳壳中3,5,6,7,8,3′,4′-七甲氧基黄酮(HMF)对湿阻中焦大鼠的改善作用。方法  将大鼠随机分为正常组、
          模型组、阳性对照组(消旋山莨菪碱片,1 mg/kg)和HMF低、中、高剂量组(0.3、0.6、0.9 mg/kg),每组7只。除正常组外,其余各组大
          鼠均采用内因加外因复合因素法造模。造模成功后,各组大鼠灌胃相应药物或生理盐水,每日1次,连续14 d。观察各组大鼠饮
          食量、饮水量及精神状态等一般行为学状态,测定大鼠粪便含水率及唾液流率。采用苏木素-伊红染色法观察大鼠胃和小肠组织
          的病理形态学变化;检测大鼠血浆中醛固酮含量;检测大鼠胃组织中水通道蛋白3(AQP3)表达水平。结果  与正常组比较,模型
          组大鼠饮食量、饮水量均显著减少(P<0.01),粪便含水率、唾液流率、血浆中醛固酮含量和胃组织中AQP3表达水平均显著增加/
          升高(P<0.01);大鼠胃组织损伤侵及黏膜肌层,黏膜肌层断裂;小肠出现少量肠绒毛糜烂,腺体结构破坏等病理形态学变化。与
          模型组比较,HMF各剂量组大鼠饮食量、饮水量均增加,粪便含水率、唾液流率、血浆中醛固酮含量和胃组织中AQP3表达水平均
          减少/降低,上述差异大部分有统计学意义(P<0.05或P<0.01);胃和小肠组织的病理形态学变化均有不同程度改善。结论  枳壳
          燥性成分HMF对湿阻中焦大鼠症状有一定的改善作用,其机制可能与降低血浆醛固酮含量、下调胃组织中AQP3表达有关。

          ABSTRACT   OBJECTIVE To investigate the improvement effects of 3,5,6,7,8,3′,4′-heptamethoxyflavone (HMF) of Fructus
          Aurantii  on  rats  with  damp  blockage  of  the  middle  energizer.  METHODS  The  rats  were  randomly  divided  into  normal  group,
          model  group,  positive  control  group (Raceanisodamine  tablet,  1  mg/kg),  HMF  low-dose,  medium-dose  and  high-dose  groups
         (0.3,  0.6,  0.9  mg/kg),  with  7  rats  in  each  group.  Except  for  the  normal  group,  the  other  groups  were  modeled  by  internal  and
          external  composite  factors. After  successful  modeling,  the  rats  in  each  group  were  given  the  corresponding  drug  or  normal  saline,
          once  a  day,  for  14  days.  The  general  behavioral  states  such  as  dietary  intake,  water  intake  and  mental  state  of  the  rats  were
          observed,  and  the  fecal  water  content  rate  and  saliva  flow  rate  were  measured.  Hematoxylin-eosin (HE)  staining  was  used  to
          observe  the  pathological  and  morphology  in  gastric  and  small  intestinal  tissues  of  rats.  The  plasma  content  of  aldosterone  was
          detected,  and  the  expression  of  aquaporins (AQP3)  in  the  gastric  tissue  of  rats  was  determined.  RESULTS  Compared  with  the
          normal group, the dietary intake and water intake of the model group rats were significantly decreased (P<0.01), the fecal water
          content  rate,  salivary  flow  rate,  plasma  content  of  aldosterone  and  the  expression  of  AQP3  in  gastric  tissue  were  increased
          significantly (P<0.01).  Gastric  tissue  injury  invaded  the  mucosal  muscle  layer,  resulting  in  mucosal  muscle  layer  rupture;
