当归多糖改善先兆流产模型大鼠Th1/Th2失衡及保胎作用                                                             Δ

          摘   要  目的  研究当归多糖通过调节磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(AKT)信号通路对先兆流产模型大鼠的保胎作用及
          Th1/Th2平衡的影响。方法  将妊娠大鼠随机分为对照组、模型组、当归多糖组(200 mg/kg)、PI3K/AKT信号通路抑制剂LY294002
          组(5 mg/kg)和当归多糖+LY294002组(200 mg/kg当归多糖+5 mg/kg LY294002),每组10只。除对照组外其余各组大鼠均灌胃米非
          司酮(8.3 mg/kg)和米索前列醇(100 μg/kg)建立先兆流产模型,并灌胃或腹腔注射相应药物。检测大鼠血清中雌激素、孕激素、干扰
          素γ(IFN-γ)、肿瘤坏死因子α(TNF-α)、白细胞介素4(IL-4)水平;测定大鼠子宫卵巢指数及胚胎死亡率;观察大鼠子宫组织形态;
          检测大鼠外周血中Th1/Th2平衡以及子宫组织中PI3K/AKT信号通路相关蛋白的表达。结果  与对照组比较,模型组大鼠子宫组
          织出现病理损伤;血清中雌激素、孕激素、IL-4水平,子宫卵巢指数、外周血中Th2细胞比例,子宫组织中p-PI3K/PI3K、p-AKT/AKT比
          值均显著降低(P<0.05);胚胎死亡率、Th1细胞比例、Th1/Th2比值和血清中IFN-γ、TNF-α水平均显著升高(P<0.05)。与模型组
          比较,当归多糖组大鼠子宫组织病理损伤减轻,上述指标均显著改善(P<0.05),且LY294002可减弱当归多糖对模型大鼠的改善
          作用(P<0.05)。结论  当归多糖可通过激活PI3K/AKT信号通路改善先兆流产模型大鼠Th1/Th2失衡,进而抑制免疫炎症,促使
          胚胎存活。

          ABSTRACT    OBJECTIVE  To  study  the  tocolysis  effects  of Angelica  sinensis  polysaccharides  on  threatened  abortion  model  rats
          and  their  impacts  on  Th1/Th2  balance  by  regulating  the  phosphoinositide  3-kinase (PI3K)/protein  kinase  B (AKT)  signaling
          pathway. METHODS Pregnant rats were randomly grouped into the control group, model group, A. sinensis polysaccharide group
         (200 mg/kg), PI3K/AKT signaling pathway inhibitor LY294002 group (5 mg/kg), and A. sinensis polysaccharide+LY294002 group
         (200  mg/kg A.  sinensis  polysaccharide+5  mg/kg  LY294002),  with  10  rats  in  each  group.  Except  for  the  control  group,  rats  in  all
          other  groups  were  given  mifepristone (8.3  mg/kg)  and  misoprostol (100  μg/kg)  intragastrically  to  establish  a  threatened  abortion
          model, and intragastric or intraperitoneal injection of corresponding drugs. The serum levels of estrogen, progesterone, interferon-γ
         (IFN-γ), tumor necrosis factor-α (TNF-α), and interleukin-4 (IL-4) in each group of rats were detected, and the uterine ovarian
          index and embryonic mortality rate of rats in each group were measured; the morphology of uterine tissue in rats was observed in
          each group; Th1/Th2 balance in peripheral blood of rats as well as the expression of PI3K/AKT signaling pathway-related proteins
                                                              in  the  uterine  tissues  of  rats  in  each  group  were  detected.
