香青兰有效部位的成分鉴定、指纹图谱建立及含量测定

摘 要 目的 为香青兰有效部位(EPDM)的全面质量评价和控制提供依据。方法 制备10批EPDM,利用高效液相色谱串联四
极杆-静电场轨道阱高分辨质谱(HPLC-Q-Exactive-MS)联用技术采集EPDM化学信息,结合文献检索、数据库比对、人工解析等手
段进行成分鉴定。根据《中药色谱指纹图谱相似度评价系统(2004A版)》建立10批EPDM的HPLC指纹图谱,进行相似度评价和
共有峰指认,并采用HPLC法测定5个指标成分的含量。结果 从EPDM中共鉴定出11个化合物。10批EPDM的指纹图谱相似度
均在 0.96 以上。从 11 个共有峰中指认出了木犀草素-7-O-β-D-葡萄糖醛酸苷(LG)、芹菜素-7-O-葡萄糖醛酸苷(APG)、迷迭香酸
(RA)、香叶木素-7-O-β-D-葡萄糖醛酸苷(DG)、田蓟苷(TL)5个色谱峰。定量分析结果显示,上述5个成分在检测质量浓度范围内
均线性良好(R ≥0.999),平均加样回收率为95.12%~98.37%;各成分含量分别为21.268 3~29.243 9 mg/g(LG)、6.365 4~7.771 7

mg/g(APG)、37.327 4~45.671 2 mg/g(RA)、17.169 9~21.985 9 mg/g(DG)、66.940 4~91.206 3 mg/g(TL)。结论 所建立的成分鉴
定、指纹图谱和含量测定方法稳定可行、结果可靠,适用于EPDM的综合质量评价和控制。

ABSTRACT OBJECTIVE To provide a reference for comprehensive quality evaluation and control of the effective parts of
Dracocephalum moldavica (EPDM). METHODS A total of 10 batches of EPDM were prepared, and chemical information of
EPDM was collected by HPLC-Q-Exactive-MS. EPDM components were identified by literature search, database comparison and
manual analysis. HPLC fingerprints of 10 batches of EPDM were established by using the Similarity Evaluation System for
Traditional Chinese Medicine Chromatographic Fingerprint (2004 A edition); the similarity evaluation and common peak
identification were carried out, and the contents of 5 index components were determined by HPLC. RESULTS A total of 11
compounds in EPDM were identified. The fingerprint similarities of EPDM samples from 10 batches were all above 0.96. Among 11
chromatographic peaks, 5 peaks were identified, such as luteolin-7-O-β-D-glucuronide(LG), apigenin-7-O-glucuronide(APG),
rosmarinic acid(RA), diosmetin-7-O-β-D-glucuronide(DG), tilianin(TL) . The results of the quantitative analysis showed that all
above 5 components had good linearity (R ≥0.999), and the average recoveries were in the range of 95.12%-98.37%. The contents

of LG, APG, RA, DG, TL were 21.268 3-29.243 9, 6.365 4-7.771 7, 37.327 4-45.671 2, 17.169 9-21.985 9, 66.940 4-91.206 3
mg/g, respectively. CONCLUSIONS The established method of component identification, fingerprint and content determination is
stable, feasible and reliable, which is suitable for the comprehensive quality evaluation and control of EPDM.
