UPLC-QAMS法同时测定连翘花中5种成分的含量

          摘   要  目的  建立同时测定连翘花中芦丁、连翘酯苷A、(+)-松脂素-4-O-β-D-吡喃葡萄糖苷、连翘苷、连翘脂素5种成分的方法。
          方法  采用超高效液相色谱(UPLC)法进行检测。色谱柱为 ACQUITY UPLC HSS T3 C18;流动相为乙腈-0.1% 磷酸溶液(梯度洗
          脱);流速为 0.3 mL/min ;检测波长为275 nm(0~8 min)、330 nm(8~10.5 min)、275 nm(10.5~32 min);柱温为25 ℃;进样量为1
          μL。以芦丁为参照物,采用一测多评(QAMS)法计算各成分含量,并与外标法进行比较。结果  QAMS法所得连翘花中连翘酯苷
          A、(+)-松脂素-4-O-β-D-吡喃葡萄糖苷、连翘苷、连翘脂素的含量分别为 7.472~7.671、2.919~2.986、1.439~1.486、1.523~1.566
          mg/g,外标法测定结果为 7.454~7.664、2.913~2.996、1.444~1.484、1.519~1.562 mg/g,两种方法测定结果无明显差异,相对偏
          差<1.0%。结论  本研究成功建立了 UPLC-QAMS 法同时测定连翘花中 5 种成分的含量,且该方法与外标法所得结果无明显差
          异,可用于连翘花的质量控制。

          ABSTRACT    OBJECTIVE To establish the methods for simultaneous determination of rutin, forsythiaside A, (+)-pinoresinol-4-
          O-β-D-glucopyranoside,  forsythin  and  forsythigenin  in  Forsythia  suspensa  flower.  METHODS  UPLC  method  was  adopted.  The
          determination was performed on ACQUITY UPLC HSS T3 C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric
          acid solution (gradient elution) at the flow rate of 0.3 mL/min. The detection wavelengths were set at 275 nm (0-8 min),330 nm
         (8-10.5 min),275 nm (10.5-32 min), respectively. The column temperature was 25 ℃, and sample size was 1 μL. Taking rutin as
          reference,  the  content  of  each  component  was  determined  by  quantitative  analysis  of  multi-components  by  single-marker (QAMS)
          method, and then compared with external standard method. RESULTS The contents of forsythiaside A, (+)-pinoresinol-4-O-β-D-
          glucopyranoside,  forsythin  and  forsythigenin  by  QAMS  were  7.472-7.671,  2.919-2.986,  1.439-1.486,  1.523-1.566  mg/g;  the
          results  obtained  by  the  external  standard  method  were  7.454-7.664,  2.913-2.996,  1.444-1.484,  1.519-1.562  mg/g,  respectively.
          There was  no significant difference in the measurement results between the two methods, with a relative deviation less than 1.0%.
          CONCLUSIONS  This  study  successfully  establishes  the  UPLC-QAMS  method  for  simultaneous  determination  of  five  components
          in  F.  suspensa  flower,  and  the  results  obtained  by  this  method  are  not  significantly  different  from  those  obtained  by  the  external
          standard method. It can be used for quality control of F. suspensa flower.

              连 翘 花 为 木 犀 科 植 物 连 翘 Forsythia  suspensa       翘、沁源连翘、陵川连翘等具有地理标志的道地药材。
         (Thunb.)Vahl的干燥花 ,具有清热解毒、消肿止痛的功                      连翘于每年3-4月先长叶,后再开花,由于花期长、花量

          效。山西是连翘的道地产地,在晋南及晋东南地区已形                            大,在其生长期需进行疏花以增加坐果率,但也因此导
          成了规模化种植,先后有安泽连翘、绛县连翘、平顺连                            致大量的连翘花被舍弃,造成资源浪费。根据文献报
