粉防己碱对神经母细胞瘤细胞增殖和迁移、侵袭能力的影响及其

        机制研究

        摘  要   目的:探讨粉防己碱(TET)对神经母细胞瘤细胞增殖和迁移、侵袭能力的影响及其作用机制。方法:以人神经母细胞瘤
        细胞系IMR-32细胞、SH-SY5Y细胞为研究对象,采用四甲基偶氮唑蓝(MTT)方法检测浓度分别为0(空白对照)、2.5、5、10、15、20
        μmol/L的TET对细胞增殖的影响;采用Transwell小室迁移实验和侵袭实验,考察正常对照组(TET浓度为0 μmol/L)和TET组(10
        μmol/L,IMR-32细胞;15 μmol/L,SH-SY5Y细胞)的细胞跨膜情况;采用Western blotting法检测按上述浓度TET处理后的IMR-32
        细胞和SH-SY5Y细胞中金属基质蛋白酶2(MMP-2)、MMP-9、β-catenin、糖原合成激酶3β(GSK3β)、磷酸化GSK3β(p-GSK3β)的
        蛋白表达水平。结果:5、10、15、20 μmol/L的TET可显著降低IMR-32细胞和SH-SY5Y细胞的存活率(P<0.05或P<0.01),其半
        数抑制浓度分别为10.148、14.461 μmol/L。细胞迁移实验和侵袭实验结果显示,与正常对照组比较,TET组跨膜细胞数均显著减
        少(P<0.01);Western blotting法检测结果显示,与正常对照组比较,TET组细胞中MMP-2、MMP-9、β-catenin、p-GSK3β蛋白表达均
        显著降低(P<0.01)。结论:TET 对神经母细胞瘤细胞的增殖和迁移、侵袭能力具有明显的抑制作用,其机制与下调细胞中
        MMP-2、MMP-9蛋白表达及抑制Wnt/β-catenin信号通路有关。
