补肾宁神膏的指纹图谱建立、含量测定及体外抗氧化作用的谱效

          摘  要  目的  建立补肾宁神膏的指纹图谱,测定其指标成分的含量,并考察其体外抗氧化活性。方法  建立10批补肾宁神膏的
          高效液相色谱(HPLC)指纹图谱,并进行相似度评价和共有峰指认;同法测定其中红景天苷等10个成分的含量;以2,2′-联氨-(3-
          乙基苯并噻唑啉-6-磺酸)二氨盐(ABTS)和1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率以及铁离子还原能力(FRAP)为指标
          考察其抗氧化活性,采用SIMCA 14.1软件进行灰色关联度分析和偏最小二乘回归分析以建立谱效关系。结果  10批补肾宁神膏
          共有24个共有峰,相似度均大于0.96;指认了11个共有峰,分别为腺苷(峰1)、红景天苷(峰4)、莫诺苷(峰6)、儿茶素(峰7)、芍药
          苷(峰10)、斯皮诺素(峰11)、阿魏酸(峰12)、异槲皮苷(峰13)、毛蕊花糖苷(峰14)、丹皮酚(峰23)及大黄素(峰24)。含量测定结
          果显示,红景天苷、莫诺苷、儿茶素、芍药苷、斯皮诺素、阿魏酸、异槲皮苷、毛蕊花糖苷、丹皮酚及大黄素的平均含量分别为0.725、
          1.962、0.214、3.395、0.124、0.107、0.286、0.019、0.034、0.067 mg/g。10批补肾宁神膏的平均综合指数为85.08%~96.35%。谱效关系
          分析结果显示,24个共有峰与其抗氧化能力均呈正相关,其中17个共有峰的变量重要性投影值>1,分别为峰2、5~7、9、10、13~
          21、23、24。结论  本研究成功建立了补肾宁神膏的HPLC指纹图谱及含量测定方法;莫诺苷等17个共有峰代表的化合物可能是其
          发挥抗氧化作用的药效成分。

          ABSTRACT   OBJECTIVE  To  establish  the  fingerprint  of  Bushen  ningshen  ointment,  determine  the  contents  of  its  major
          constituents,  and  investigate  its  in  vitro  antioxidant  activity.  METHODS  High  performance  liquid  chromatography (HPLC)
          fingerprints  of  10  batches  of  Bushen  ningshen  ointment  were  established.  Similarity  evaluation  and  identification  of  common  peaks
          were  subsequently  performed.  The  contents  of  10  components  such  as  salidroside  were  determined  using  the  same  HPLC  method.
          Using  the  scavenging  rates  against  2,2′-azino  bis(3-ethylbenzothiazoline-6-sulfonic  acid)  (ABTS)  and  1,1-diphenyl-2-
          picrylhydrazyl (DPPH) radicals, as well as ferric ion reducing antioxidant power (FRAP) as indicators, the anti-oxidant activity of
          the ointment was evaluated; grey relational analysis and partial least squares regression were conducted using SIMCA 14.1 software
          to  establish  the  spectrum-effect  relationship.  RESULTS  The  fingerprint  chromatogram  of  10  batches  of  Bushen  ningshen  ointment
